    Assessment of alternative power sources for mobile mining machinery

    Alternative mobile power sources for mining applications were assessed. A wide variety of heat engines and energy systems was examined as potential alternatives to presently used power systems. The present mobile power systems are electrical trailing cable, electrical battery, and diesel - with diesel being largely limited in the United States to noncoal mines. Each candidate power source was evaluated for the following requirements: (1) ability to achieve the duty cycle; (2) ability to meet Government regulations; (3) availability (production readiness); (4) market availability; and (5) packaging capability. Screening reduced the list of candidates to the following power sources: diesel, stirling, gas turbine, rankine (steam), advanced electric (batteries), mechanical energy storage (flywheel), and use of hydrogen evolved from metal hydrides. This list of candidates is divided into two classes of alternative power sources for mining applications, heat engines and energy storage systems

    Experimental study of ceramic coated tip seals for turbojet engines

    Ceramic gas-path seals were fabricated and successfully operated over 1000 cycles from flight idle to maximum power in a small turboshaft engine. The seals were fabricated by plasma spraying zirconia over a NiCoCrAlX bond boat on the Haynes 25 substrate. Coolant-side substrate temperatures and related engine parameters were recorded. Post-test inspection revealed mudflat surface cracking with penetration to the ceramic bond-coat interface

    A parametric level-set method for partially discrete tomography

    This paper introduces a parametric level-set method for tomographic reconstruction of partially discrete images. Such images consist of a continuously varying background and an anomaly with a constant (known) grey-value. We represent the geometry of the anomaly using a level-set function, which we represent using radial basis functions. We pose the reconstruction problem as a bi-level optimization problem in terms of the background and coefficients for the level-set function. To constrain the background reconstruction we impose smoothness through Tikhonov regularization. The bi-level optimization problem is solved in an alternating fashion; in each iteration we first reconstruct the background and consequently update the level-set function. We test our method on numerical phantoms and show that we can successfully reconstruct the geometry of the anomaly, even from limited data. On these phantoms, our method outperforms Total Variation reconstruction, DART and P-DART.Comment: Paper submitted to 20th International Conference on Discrete Geometry for Computer Imager

    Inhibition of AMP-Activated Protein Kinase Signaling Alleviates Impairments in Hippocampal Synaptic Plasticity Induced by Amyloid beta

    The AMP-activated protein kinase (AMPK) is a Ser/Thr kinase that is activated in response to low-energy states to coordinate multiple signaling pathways to maintain cellular energy homeostasis. Dysregulation of AMPK signaling has been observed in Alzheimer\u27s disease (AD), which is associated with abnormal neuronal energy metabolism. In the current study we tested the hypothesis that aberrant AMPK signaling underlies AD-associated synaptic plasticity impairments by using pharmacological and genetic approaches. We found that amyloid beta (A beta)-induced inhibition of long-term potentiation (LTP) and enhancement of long-term depression were corrected by the AMPK inhibitor compound C (CC). Similarly, LTP impairments in APP/PS1 transgenic mice that model AD were improved by CC treatment. In addition, A beta-induced LTP failure was prevented in mice with genetic deletion of the AMPK alpha 2-subunit, the predominant AMPK catalytic subunit in the brain. Furthermore, we found that eukaryotic elongation factor 2 (eEF2) and its kinase eEF2K are key downstream effectors that mediate the detrimental effects of hyperactive AMPK in AD pathophysiology. Our findings describe a previously unrecognized role of aberrant AMPK signaling in AD-related synaptic pathophysiology and reveal a potential therapeutic target for AD

    Experimental and analytical study of ceramic-coated turbine-tip shroud seals for small turbine engines

    The results of an experimental and analytical evaluation of ceramic turbine tip shrouds within a small turbine engine operating environment are presented. The ceramic shrouds were subjected to 1001 cycles between idle and high power and steady-state conditions for a total of 57.8 engine hr. Posttest engine inspection revealed mud-flat surface cracking, which was attributed to microcracking under tension with crack penetration to the ceramic and bond coat interface. Sections and micrographs tend to corroborate the thesis. The engine test data provided input to a thermomechanical analysis to predict temperature and stress profiles throughout the ceramic gas-path seal. The analysis predicts cyclic thermal stresses large enough to cause the seal to fail. These stresses are, however, mitigated by inelastic behavior of the shroud materials and by the microfracturing that tensile stresses produce. Microfracturing enhances shroud longevity during early life but provides the failure mechanism during life but provides the failure mechanism during extended life when coupled with the time dependent inelastic materials effects

    DEVELOPMENT OF A STRATEGIC BUSINESS DECISION-MAKING ENVIRONMENT FOR COMMERCIAL JET ENGINE SELECTION

    Presented at the 41st AIAA Aerospace Sciences, Meeting and Exhibit, Reno, NV, January 6-10, 2003.In today?s business climate, aerospace companies are more than ever in need of rational methods and techniques that provide insights as to the best strategies which may be pursued for increased profitability and risk mitigation. However, the use of subjective, anecdotal decision-making remains prevalent due to the absence of analytical methods capable of capturing and forecasting future needs. Negotiations between airframe and engine manufacturers could benefit greatly from a structured environment that facilitates efficient, rational, decision-making. Creation of such an environment can be developed through a parametric physics-based, stochastic formulation that uses meta-models to expedite the process. This paper describes such an approach in order to demonstrate the types of insights that might be gained as an engine manufacturer tries to forecast the effects of uncertainties and future vehicle requirements on engine related characteristics for the design of a hypothetical regional business jet. Game theory concepts are suggested as a potential means by which one can attach business payoffs to the selection of any engine design point

    Aprotinin Inhibits SARS-CoV-2 Replication

    Severe acute respiratory syndrome virus 2 (SARS-CoV-2) is the cause of the current coronavirus disease 19 (COVID-19) pandemic. Protease inhibitors are under consideration as virus entry inhibitors that prevent the cleavage of the coronavirus spike (S) protein by cellular proteases. Herein, we showed that the protease inhibitor aprotinin (but not the protease inhibitor SERPINA1/alpha-1 antitrypsin) inhibited SARS-CoV-2 replication in therapeutically achievable concentrations. An analysis of proteomics and translatome data indicated that SARS-CoV-2 replication is associated with a downregulation of host cell protease inhibitors. Hence, aprotinin may compensate for downregulated host cell proteases during later virus replication cycles. Aprotinin displayed anti-SARS-CoV-2 activity in different cell types (Caco2, Calu-3, and primary bronchial epithelial cell air–liquid interface cultures) and against four virus isolates. In conclusion, therapeutic aprotinin concentrations exert anti-SARS-CoV-2 activity. An approved aprotinin aerosol may have potential for the early local control of SARS-CoV-2 replication and the prevention of COVID-19 progression to a severe, systemic disease
